However, an epigenetic therapy developed by the American biotechnology company Life Biosciences has radically shifted these prevailing views.

Scientists at Life Biosciences moved beyond merely temporarily halting disease symptoms and set the goal of rejuvenating the functional age of cells. To this end, they created ER-100, a revolutionary biotechnological therapy that stops the cellular aging process and seeks to fully restore damaged nervous tissue.

At the end of January of this year, Life Biosciences officially received approval from the U.S. Food and Drug Administration (FDA) to conduct clinical trials of ER-100 on humans. Following this historic FDA decision, the process moved to a practical stage, and research—aimed at assessing the safety and efficacy of the epigenetic therapy—has already begun as of March.

The study is currently in the “Not yet recruiting” phase, meaning that despite the FDA’s approval, the scientific team is still undergoing final preparatory procedures before beginning experiments on patients.

What is ER-100 and how does its mechanism work?

ER-100 is a completely new generation of biotechnology. Its mechanism of action is based on the partial reprogramming of a cell’s functional state. During the aging process, epigenetic markers—molecular changes that do not alter the DNA code but influence which genes are activated and in what sequence—accumulate in cells. Over time, these changes disrupt normal gene regulation, causing cells to gradually lose their ability to function healthily. The goal of ER-100 is to partially correct these epigenetic disruptions and restore the cell to a more youthful, functionally active state.

ER-100 corrects these disruptions through the following mechanism: by using a safe viral vector (AAV), it delivers instructions to the cell to produce three specific proteins (OSK). These proteins, in turn, clear the cell of accumulated harmful markers and return it to its original, functionally active state. It is important to note that ER-100 does not alter or damage the patient’s DNA code and does not transform the cell into a different type.

Preclinical research results

Before the start of human clinical trials for ER-100, the preclinical stage yielded convincing results, primarily based on experiments conducted on mouse models. Scientists proved that it is possible to partially reprogram the epigenetic profile of cells using OSK factors (Oct4, Sox2, Klf4).

Research confirmed that this method can lead to the regrowth of damaged optic nerve axons and effectively turn back the cellular “biological clock.” The most important discovery was that this process did not result in the development of cancerous processes, which had been considered one of the primary risks.

Ultimately, scientists focused on curing degenerative diseases that cause vision loss, specifically glaucoma. According to the experiment, the mice showed significant improvement in visual acuity and reaction to visual stimuli. All of this indicates that the rejuvenation achieved at the cellular level directly reflected in the restoration of the physiological function of the optic nerve.

These research results became the main foundation for the FDA granting Life Biosciences permission for human testing, as the hypothesis was confirmed: epigenetic therapy actually restores damaged nervous tissue.

Why was the FDA’s approval so important?

The authorization granted by the FDA to begin clinical trials is a decisive stage, confirming that the epigenetic therapy has passed rigorous preclinical safety and efficacy evaluations and is ready to be tested on humans.

This approval essentially serves as a mark of scientific credibility, as FDA standards are considered the strictest in the world. Through this mechanism, the U.S. protects patients from unproven or potentially harmful medical interventions and provides a legal basis for the practical realization of innovative projects like the one by Life Biosciences.

The importance of clinical trials and future prospects

Although preliminary data on the initial results of this biotechnology may become available by the end of 2026, the full study cycle and the evaluation of its long-term impact are scheduled for 2032.

This long-term observation is crucial to determine how sustainable the effect of epigenetic reprogramming is and whether it can stabilize the optic nerve restoration process in the human body. If the efficacy of ER-100 is confirmed, it will revolutionize medicine and lay the foundation for entirely new therapeutic approaches.

For decades, science viewed aging and its associated degenerative diseases, such as vision loss, as an inevitable condition.
