Notably, athletes also use it to manage pulmonary arterial hypertension (PAH), as the drug increases the body’s exercise capacity. A study published in Molecular Biology Reports confirms that tadalafil directly helps muscle cells proliferate and cope with stress.

As a PDE5 inhibitor, tadalafil blocks a specific enzyme, increases levels of the chemical messenger cyclic guanosine monophosphate (cGMP) inside cells, and enhances nitric oxide (NO) signaling. This mechanism improves blood circulation and activates stem cells known as satellite cells, which help repair damaged tissue. Doctors had previously considered tadalafil a potential treatment for Duchenne muscular dystrophy (DMD), but its long-term effects on cell growth remained insufficiently understood.

In laboratory experiments, scientists treated mouse muscle cells (the C2C12 model) with tadalafil. They used a concentration of the drug (1 µM) consistent with levels found in human blood during treatment and monitored the cells for seven days.

The medication markedly increased cell viability and proliferation. In a simulated wound-healing assay, the cells filled the gap within 24 hours. In addition, the drug delayed premature cellular maturation, preserving a large pool of immature cells available for tissue repair.

In cultures treated with tadalafil, the researchers detected elevated levels of early muscle development markers, including Pax7 and Myf6. The drug also increased the release of IL-6, a signaling molecule that activates stem cells. At the same time, it suppressed myostatin and TGF-β, two key proteins that inhibit muscle growth.

The researchers consider these laboratory findings promising. However, further studies, including clinical research, will be needed to establish whether tadalafil offers genuine therapeutic benefits in living organisms.

MedicalXpress

The medication tadalafil, which is used in medicine to treat erectile dysfunction and an enlarged prostate, significantly accelerates muscle tissue regeneration.
