A new study published in the journal Alzheimer’s & Dementia: The Journal of the Alzheimer’s Association has found that an impairment of the brain’s glymphatic system, which is responsible for clearing waste and toxins from the central nervous system, is linked to an increased risk of dementia.
The research, which is based on data from over 45,000 participants in the UK Biobank, confirms that a deterioration in cerebrospinal fluid (CSF) dynamics contributes to the accumulation of toxic proteins that cause dementia.
The researchers used machine learning algorithms to analyze MRI scans and identified three biomarkers that allow for the assessment of glymphatic function and the prediction of dementia risk: a low risk of dementia is associated with a high DTI-ALPS index (which indicates healthy fluid movement in the brain’s clearance channels), while the risk increases in cases of low BOLD-CSF coupling and a large volume of the CSF-producing structure (choroid plexus).
The study showed that cardiovascular factors such as hypertension, diabetes, smoking, and excessive alcohol consumption damage glymphatic function. Experts suggest that these conditions reduce arterial pulsation and impede CSF flow, leading to the accumulation of toxic waste.
The findings indicate that improving glymphatic function is a powerful tool for reducing the risk of dementia. Since glymphatic activity is highest during deep sleep, maintaining sleep hygiene and cardiovascular health is critically important.
In the future, these MRI-based biomarkers may be used for the early diagnosis of dementia, allowing doctors to prescribe timely preventative treatments.
