A decisive breakthrough in research for the treatment of HIV, the Human Immunodeficiency Virus. For years, scientists have battled one of HIV’s most enigmatic characteristics: its ability to hide within immune cells in a latent (dormant) state. Even with treatment, this “sleeping” virus reactivates as soon as therapy is stopped. Now, the “new mouse model, μ-Lat,” created by leading American research institutes, allows scientists to rapidly, effectively, and affordably test HIV cure strategies.
How μ-Lat Works and Why It’s Different
Researchers used specialized laboratory cells called J-Lat—a clone of human immune cells that contains the HIV genetic code in an inactive, “dormant” state. These cells were transferred into immunodeficient mice (NSG strain), where they successfully spread to various organs: bone marrow, spleen, lungs, and blood.
The main achievement is that the virus remained in a latent state, meaning the model realistically replicates the mechanism of HIV latency. The next step involved the targeted “awakening” of the virus. Scientists injected TNF-α—a cytokine known for its ability to activate latent HIV—into the mice.
The results proved impressive:
The virus was activated even in organs known to be difficult to study.
Using a special biomarker (GFP), the areas of viral activation were precisely determined.
For the first time, the mouse model confirmed that it is possible to test treatments to determine whether the therapy reaches the tissues and is successful in “awakening” the latent virus.
Why Is This a Breakthrough?
Previous models were expensive and complex, relying mainly on primates or difficult-to-reconstruct humanized systems. In contrast, μ-Lat is cheap, simple, accessible, and, most importantly, highly specific. This model allows scientists to assess the effectiveness of medications in the organs where HIV often hides. They can also investigate how well therapy can “flush out” the virus from those latent reservoirs that remain the greatest challenge today.
Does This Mean HIV Is Now Curable?
Not yet. However, this is a huge step. Current drugs only suppress the virus but cannot reach those “hidden reservoirs” where HIV is dormant. μ-Lat was created specifically to solve this problem, namely to study whether a particular therapy can fully remove the virus from the body.
